Inhibit IL11, improve function and prolong lifespan

IL-11 is a cytokine, a protein encoded by the IL11 gene in human use. It can regulate adipogenesis, cell proliferation, neurogenesis and pre-saturation. However, overexpressed IL-11 is clearly associated with inflammation and cancer.

In July 2024, Professor Weiwen Lin of Duke-National University of Singapore Medical School and others published a heavyweight paper in Nature, "Inhibition of IL-11 signaling can extend the healthy lifespan and lifespan of mammals", pointing out that IL-11 is a pro-inflammatory and pro-fibrotic member of the IL-6 family, which may promote age-related pathology and continue to grow. This premise is based on the proof that IL-11 can activate ERK-mTORC1 and/or JAK-STAT3. Based on.

The author believes that the population consumption of IL_11 can be seen from many aspects, including IL-11 upregulation with age; inducing cell aging; related aging, etc. (1)

In further experiments, they found that the mouse model without IL-11 could significantly improve function, and the degree of weakness and fibrosis, gait and many other indicators were better than wild mice; especially in terms of power consumption, after the IL-11 mouse model, the power consumption of female wild-type mice was 120.9 weeks, and that of IL-11 mice was 148.3 weeks, an increase of 22%; the life span of closed wild mice was 130.3 weeks, and after IL-11 inhibition, it was 159.6 weeks, an increase of 22.5%, which fully proved the assertion that inhibiting IL-11 can prolong life.

Scientists from the University of Singapore emphasized that trametinib, rapamycin or metformin can prolong the life span of model organisms by inhibiting ERK or mTOR and activating AMPK, respectively, and some people use such drugs in humans. However, these drugs have on-target and off-target toxicity, as well as variable effects on health doses and factors, which are sometimes harmful. Our data show that anti-IL-11 treatment has a reassuring safety and can be used to extend human healthy life span through potential translational methods. (1)

Medical research, especially in mouse experiments, inhibits IL11 using antibodies with high concentrations, such as X203, X209, etc. Since last year, new antibodies have been developed, with more types and direct effects. For example, the IL-11R¦Á antibody LASN-01 developed by Lassen is a highly targeted fully human monoclonal antibody that can bind to IL-11R with high affinity and inhibit IL-11 signal closely. (Bai Ying Biology)
However, it is impossible to use such things in the process of personal anti-aging and life extension. The drug concentration and drug efficacy are directly related, similar to the inhibition of mTORC1. Exceeding the regulation will increase S6K activity and eternal life. DDC operation, so insisting on low doses is to consider the balance between dose, effect and safety. Among our supplements, there are seven or eight supplements that inhibit IL11, such as flavin, astaxanthin, tetracortin, rutin, Omega-3, taurine, etc. There are basically no obvious reactions when checking these supplements used in the market. As for the effect, it is speculated that it should be mild and powerful.
